A car fire is revealed to be the cause of the second police incident on Auchinlea Road.
Emergency services remain on the scene of the incident.
The road is closed from Auchinlea Way, which is used to access Glasgow Fort and Coinsborough Road.
The road was first closed in both directions around 3pm on Tuesday due to a separate police incident.
It reopened shortly after.

Police informed the Glasgow Times the first incident was due to a car crash.
Auchinlea Road closed for a second time around 5pm.
A Traffcom bulletin revealed the police incident was related to a car fire.
Drivers are asked to find an alternate route if possible.
The Traffcom bulletin read: "Due to an ongoing Police Incident involving a car fire, Auchinlea Road is closed from Auchinlea Way (Access to Glasgow Fort) and Coinsborough Road.
"Emergency Services are in attendance.
"Drivers should seek an alternative route if possible."
The road has since fully reopened.
